DNA replication uses the enzyme DNA polymerase in pairing up the bases from the template strand to the extending complementary strand. It uses the primers as the starting point of replication. This enzyme, however, is limited to synthesis of polynucleotides from the 5' to 3' direction. Therefore, base pairing would always start at the 3' side of the template strand.
